    Keratoacanthoma and keratoacanthoma-like squamous cell carcinoma similar morphology but different pathogenesis

The mechanisms that drive their distinct biological behavior are poorly understood.To investigate whether the assessment of microvessel density (MVD) could be helpful in KA and SCC differential diagnosis and to gain insight into the pathogenesis of KA-like neoplasms, we compared the density of CD105- and CD34-stained vessels in KAs and SCCs and their relation to the expression of the p53 oncoprotein and proliferation marker Ki67.This is an observational retrospective cohort study. Forty lesions with clinical appearance of KAs (29 KAs and 11 SCCs) entered the study. A biopsy was taken from each lesion at presentation and the natural clinical course was monitored for at least 1 month. Growing or minimally regressing lesions were submitted to complete surgical excision. The diagnoses were established on combined clinical, histological, and follow-up evaluations. The MVD and p53 or Ki67 expression in neoplastic cells were assessed through morphometry.The MVD did not show discriminating power between KAs and SCCs. The Ki67 proliferation rate was significantly higher in SCCs. Although neoangiogenesis (CD105-MVD) in KAs was associated with cell proliferation, in SCCs it was not. There was significant correlation between p53 expression and neoplasia size in SCCs but not in KAs.From our results, we may conclude that KA and SCC have similarities, as CD105- and CD34-MVD. However, the low Ki67 proliferation index and the positive correlation between Ki-67 index and neovascularization in KA suggest a dependence in neovascularization to grow in KA, pointing to involvement of distinct pathogenesis.Differential diagnosis between keratoacanthoma (KA) and squamous cell carcinoma (SCC) is difficult due to their similarities.     Assessing local acid mine drainage impacts on natural regeneration-revegetation of São Domingos mine (Portugal) using a mineralogical, biochemical and textural approach

    São Domingos sulfide mine was shut down more than 50 years ago leaving behind eroded and depositional surfaces due to acid mine drainage (AMD). The aim of this study was to assess six selected sites subjected to AMD, considered phytotoxic regions characterized by vegetation scarcity. Two main criteria, nature and composition of soluble fractions and total chemistry of surficial products related to jarosites presence, enabled to set up an overall dichotomy between superficial proximal/discharge and distal/sedimentation areas. Wet and dry sieving results comparison revealed that samples have a predominant sandy texture and lithic (phyllite, quartzite and volcanic country rocks) composition. Quartz, and subordinate feldspar enrichment is also detected in the coarse silt fraction. The results also suggest that the materials under study, when subjected to the local torrential hydrologic regime, have a high mechanical vulnerability, facilitating erosion and mud transport, both critical for vegetation support, and triggering contamination transfer and dispersion. The vicinity and ground-level surfaces of discharging areas are enriched in the jarosite group minerals whereas the sedimentation ones present hypersaline aluminous tendency. The formation of jarosite is considered as an efficient positive environmental contribution to metals and metalloids sequestration/immobilization. The remediation/revegetation solutions to be adopted in each location must have into consideration these differentiating aspects

    Microglia dysfunction caused by the loss of Rhoa disrupts neuronal physiology and leads to neurodegeneration

Using conditional gene targeting in mice, we demonstrate that genetic ablation of the small GTPase Rhoa in adult microglia is sufficient to trigger spontaneous microglia activation, producing a neurological phenotype (including synapse and neuron loss, impairment of long-term potentiation [LTP], formation of β-amyloid plaques, and memory deficits). Mechanistically, loss of Rhoa in microglia triggers Src activation and Src-mediated tumor necrosis factor (TNF) production, leading to excitotoxic glutamate secretion. Inhibiting Src in microglia Rhoa-deficient mice attenuates microglia dysregulation and the ensuing neurological phenotype. We also find that the Rhoa/Src signaling pathway is disrupted in microglia of the APP/PS1 mouse model of Alzheimer disease and that low doses of Aβ oligomers trigger microglia neurotoxic polarization through the disruption of Rhoa-to-Src signaling.     Seasonality Role on the Phenolics from Cultivated Baccharis dracunculifolia

    Baccharis dracunculifolia is the source of Brazilian green propolis (BGP). Considering the broad spectrum of biological activities attributed to green proplis, B. dracunculifolia has a great potential for the development of new cosmetic and pharmaceutical products. In this work, the cultivation of 10 different populations of native B. dracunculifolia had been undertaken aiming to determine the role of seasonality on its phenolic compounds. For this purpose, fruits of this plant were collected from populations of 10 different regions, and 100 individuals of each population were cultivated in an experimental area of 1800 m2. With respect to cultivation, the yields of dry plant, essential oil and crude extract were measured monthly resulting in mean values of 399 ± 80 g, 0.6 ± 0.1% and 20 ± 4%, respectively. The HPLC analysis allowed detecting seven phenolic compounds: caffeic acid, ferulic acid, aromadendrin-4′-methyl ether (AME), isosakuranetin, artepillin C, baccharin and 2-dimethyl-6-carboxyethenyl-2H-1-benzopyran acid, which were the major ones throughout the 1-year monthly analysis. Caffeic acid was detected in all cultivated populations with mean of 4.0%. AME displayed the wide variation in relation to other compounds showing means values of 0.65 ± 0.13% at last quarter. Isosakuranetin and artepillin C showed increasing concentrations with values between 0% and 1.4% and 0% and 1.09%, respectively. The obtained results allow suggesting that the best time for harvesting this plant, in order to obtain good qualitative and quantitative results for these phenolic compounds, is between December and April

    Preliminary identification of the bioremediation limiting factors of a clay bearing soil contaminated with crude oil

    Bioremediation is an attractive alternative to treat soils contaminated with petroleum hydrocarbons. However, the effectiveness of biodegradation process can be limited by both contaminant characteristics and its bioavailability in soil. This work aims at establishing a preliminary procedure to identify the main factor (hydrocarbon recalcitrance or its bioavailability) that impairs the biodegradation, possibly resulting in low remediation efficiencies. Tests in soil microcosms were carried out applying biostimulation and bioaugmentation techniques. Similar experiments were conducted using liquid medium containing crude oil and microorganisms, both of them extracted from a weathered contaminated soil sample, nutrients and microbial inoculum constituted of Rhodoturula glutinis and Nocardia nova. After 30 days, highest microcosms biodegradation levels (98.8%) were obtained with bioaugmentation in liquid medium, pointing out that the main problem of petroleum hydrocarbon utilization by microorganisms was associated to its availability and not only to the intrinsic recalcitrancy of remaining pollutants

    Revisão do Conhecimento sobre os Mamíferos Aquáticos da Costa Norte do Brasil

    The Brazilian north coast represents a new frontier in several fields of knowledge. The aquatic mammals of the Brazilian northern coast were poorly studied until recently. Recent efforts promoted by Projeto Piatam Oceano are changing the scenario. New research groups on aquatic mammals were formed and are conducting a large survey of this unique group of mammals. A total of 22 species of aquatic mammals were recorded along the Brazilian northern region. It should be emphasized the rediscovery of the Antillean manatee in several localities along the coasts of Amapá, Pará and Maranhão states. On the other hand, the estuarine dolphin is widely distributed along the Amazonian coast but is frequently by-caught in gill nets fisheries throughout the northern coast. The intense use of love charms has also a negative effect on these populations and should be better evaluated. An awareness program will be implemented as a first step for changing attitude towards the aquatic mammals of the northern coast.A costa norte do Brasil representa uma nova fronteira para o avanço do conhecimento sobre a fauna de mamíferos aquáticos. A ausência de dados pretéritos sobre os mamíferos aquáticos nessa região e a própria dificuldade logística de monitoramento resultou, até muito recentemente, numa lacuna de conhecimento. A fim de reverter este quadro, o Projeto Piatam Oceano vem incentivando a formação de grupos de pesquisa sobre mamíferos aquáticos da costa amazônica. Como resultado dos esforços recentes, foi reunido um conjunto relevante de informações inéditas ou complementares sobre as 22 espécies de mamíferos aquáticos registradas até o momento na região. Destacam-se as áreas de ocorrência do peixe-boi-marinho no litoral dos Estados do Maranhão, Pará e Amapá, as quais poderiam garantir a manutenção destas populações criticamente ameaçadas. Merece destaque ainda a ocorrência do boto-cinza em toda costa amazônica e sua problemática de interação com a pesca artesanal e o comércio de peças anatômicas. Sugere-se uma ampla campanha de conscientização e a promoção de campanhas educativas visando uma nova tomada de atitude em relação aos mamíferos aquáticos e seus hábitats, respeitando os valores tradicionais e o conhecimento etnoecológico das comunidades costeiras

    Integrative overview of the herpetofauna from serra da mocidade, a granitic mountain range in northern Brazil

    The Brazilian mountain ranges from the Guiana Shield highlands are largely unexplored, with an understudied herpetofauna. Here the amphibian and reptile species diversity of the remote Serra da Mocidade mountain range, located in extreme northern Brazil, is reported upon, and biogeographical affinities and taxonomic highlights are discussed. A 22-days expedition to this mountain range was undertaken during which specimens were sampled at four distinct altitudinal levels (600, 960, 1,060 and 1,365 m above sea level) using six complementary methods. Specimens were identified through an integrated approach that considered morphological, bioacoustical, and molecular analyses. Fifty-one species (23 amphibians and 28 reptiles) were found, a comparable richness to other mountain ranges in the region. The recorded assemblage showed a mixed compositional influence from assemblages typical of other mountain ranges and lowland forest habitats in the region. Most of the taxa occupying the Serra da Mocidade mountain range are typical of the Guiana Shield or widely distributed in the Amazon. Extensions of known distribution ranges and candidate undescribed taxa are also recorded. This is the first herpetofaunal expedition that accessed the higher altitudinal levels of this mountain range, contributing to the basic knowledge of these groups in remote areas. © Leandro J.C.L.
